Molecular screening, isolation and characterization of conjugative plasmids from contaminated soils

Objectives and individual tasks of the joint project:Work carried out in India:

  1. Screening for both conjugative plasmids from gram positive bacteria and gram negative bacteria from highly contaminated soil by the triparental mating method.
  2. Isolation and incompatibility group/oriT-family-based characterization of conjugative plasmids from gram positive and gram negative bacteria from differently contaminated soils from various origins.

Work carried out in Germany:

  1. Design of a gfp-labelled pIP501-based mobilizable plasmid to capture conjugative plasmids from differently contaminated Indian and German soil samples.
  2. Establishment of a biofilm reactor system with bacterial isolates from different soil samples and plasmid transfer experiments with gfp-labelled pIP501 derivative
  3. Establishment of biofilm reactors with contaminated soil as carrier material from both India and Germany and plasmid transfer experiments with gfp-labelled pIP501 derivative
