Our aim is to elucidate the role of the
protein components in the assembly of a conjugative T4S-like machinery
in Gram-positive bacteria. We will verify our in vivo and in vitro
protein-protein interaction studies and computer predictions by
localization of the T4SLS proteins in Enterococcus faecalis cell
compartments by cell fractionation followed by Western Blot,
immunogold transmission electron microscopy and immunofluorescence
microscopy.

Objectives and individual tasks of the joint
project:Work carried out in India: Screening for both conjugative
plasmids from gram positive bacteria and gram negative bacteria from
highly contaminated soil by the triparental mating method. Isolation
and incompatibility group/oriT-family-based characterization of
conjugative plasmids from gram positive and gram negative bacteria
from differently contaminated soils from various origins

Increasing resistance of bacteria to
commonly used antibiotics has become a serious problem. Food bacteria
could act as reservoirs for resistance genes. There are very few
systematic studies on antibiotic resistant lactic acid bacteria (LAB)
in traditional fermented dairy and meat products. The project focuses
on monitoring of antibiotic resistance in LAB isolates from Turkish
dairy and meat products, as well as on the characterisation of
resistance and transfer (tra) genes carried by them. Main objectives
are the identification of lactic acid bacteria to species level in
fermented Turkish food products, determination of their inherent
antibiotic resistance and their horizontal transmissibility.

COMICS [12]
The COMICS project (Concordia Microbial
Dynamics) based on the MISSEX project focuses on analysis of the
microbial diversity of the bacterial populations from the Antarctica
Research Station CONCORDIA. Microbial diversity will be analysed on
crew samples and on samples from the technical equipment of the
research station. Emphasis is laid on Gram-positive (G+) nosocomial
and pathogenic bacteria and their plasmids. Plasmids will be analysed
with respect to size, conformation, DNA transfer and mobilization
abilities.
